Formalin-fixed paraffin-embedded tissue specimens, often called FFPE samples, are a fantastic material for understanding human biology and disease pathways. How are FFPE samples prepared?

The extraction of specimens is mainly done during biopsies and clinical and surgical procedures by a trained medical doctor or surgeon. The excised human samples are immersed in 10% neutral buffered formalin for at least 24 hours. This process is known as formalin fixation. Later, the fixed samples are dehydrated with ethanol concentrates. The tissue is then embedded in paraffin wax to create FFPE blocks, which preserve the proteins and tissue structures. Finally, these FFPE blocks are sectioned into specific sizes to prepare FFPE specimens that are investigated and authorized by a certified histopathologist for research use. FFPE samples in the form of FFPE blocks, curls, and slides can be stored for long years without degradation. Moreover, FFPE is stored at room temperature and thus does not require highly sophisticated and expensive tissue preservation units.
